DNA Purification - Wizard Columns

1. Reduce PCR reaction to 100 mL in volume.

2. Add 100 µL Wizard Buffer and mix.

3. Add 1 µL Wizard Slurry with open-bore blue tip.

4. Vortex for 5 sec. Allow to stand 15 sec. Repeat twice more.

5. Insert 3 mL syringe into Wizard Column. Place on top of a 6 mL disposable test tube.

6. Add Wizard slurry to syringe. Force slurry onto column at a rate of 1 drop/sec.

7. Add 2 mL 80% Isopropanol to the syringe. Force over column at a similar rate to rinse.

8. Remove column in place in 1.5 mL microcentrifuge tube carrier. Spin for 60 sec.

9. Discard effluent and add 50 µL sterile water directly to the column. Allow to stand 3 min.

10. Place column in a sterile 1.5 mL microcentrifuge tube carrier. Spin for 60 sec.

11. Discard column and save effluent.
