Preparation of DEAE Nitrocellulose Membranes

1. Purchase Scleicher and Schull N-45 DEAE Ion exchange paper.

2. Prepare membrane by cutting into conveniently sized portions and rinsing as follows:

	1. Rinse for 5 min in 10 mM EDTA, pH 8.0
	2. Rinse for 10 min in 0.5 M NaOH
	3. Rinse for 2 min in sterile water.
	4. Repeat water rinses 6 times.
	5. Store paper in water at 4°.

3. Prepare:

	Low Salt Buffer
	5 mL 	1 M Tris pH 8.0
	3 mL	5 M NaCl
	2 mL	500 mM EDTA, pH 8.0
	90 mL	ddH₂O

	High Salt Buffer
	5 mL 	1 M Tris pH 8.0
	20 mL	5 M NaCl
	2 mL	500 mM EDTA, pH 8.0
	73 mL	ddH₂O
